Anti-Human Secondary Antibodies

The Thermo Scientific Pierce Anti-Human Secondary Antibodies are affinity-purified polyclonal antibodies with well-characterized specificity for human immunoglobulin classes, subclasses, and fragments. They are available as unlabeled antibodies as well as conjugates of alkaline phosphatase (AP), horseradish peroxidase (HRP), Dylight fluorescent conjugates, and biotin. Host species include goat, rabbit, mouse, chicken, cow, rat and sheep.

Thermo Scientific Pierce secondary antibodies are available as whole IgG or the F(ab')2 fragment. Each of these antibodies is affinity purified using its target antigen (the primary antibody class, subclass, and fragment) coupled to an agarose support. This affinity purification eliminates nonspecific antibodies, resulting in high specificity and low background signal. Conjugated antibodies are affinity purified before labeling.

Selected secondary antibodies have been further purified by adsorbing the antigen-purified antibody to an affinity column containing immobilized serum proteins of selected species. This additional processing minimizes antibody cross-reactivity to the serum proteins of those selected species. These antibodies are designated as Cross Adsorbed.

    Western blot analysis of humanized mouse Fab was performed by loading 100ug of L3.6pL pancreatic cancer cell lysate spiked with 2ug of humanized Fab (left lanes) or 100ug of L3.6pL cell lysate (right lanes) per well onto an SDS-PAGE gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% milk in PBST for 4 hours. The membrane was probed with an anti-GAPDH mouse monoclonal antibody followed by an HRP-conjugated goat anti-human IgG F(ab’)2 secondary antibody (Product # 31482) at a dilution of 1:10,000 for 1 hour and DAB substrate (Panel A). Because GAPDH was not detected with Product # 31482, the blot was rinsed with PBS and probed with an anti-mouse IgG-HRP secondary antibody diluted 1:4000 follwed by DAB substrate (Panel B). Detection was performed using 1-Step Ultra TMB substrate (Product # 34028) for 5 minutes at room temperature in the dark. The reaction was stopped with 0.16M sulfuric acid, and absorbances were read on a spectrophotometer at 450-550nm.

IgA heavy chain was detected at ~55kD using a mouse anti-human IgA monoclonal antibody (Product # MA5-11208) diluted to a concentration of 0.5ug/ml in blocking buffer, overnight at 4C overnight on a rocking platform. The blot was washed in TBST, and probed with an HRP-conjugated goat anti-mouse light chain secondary antibody at a dilution of 1:40,000 for at least 30 minutes at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico.

To verify the presence of all isotypes, a second blot was probed with a goat anti-human IgM + IgG +IgA polyclonal antibody (Product # 31128) at a concentration of 0.5ug/ml in blocking buffer overnight at 4C on a rocking platform, followed by an HRP-conjugated mouse anti-goat IgG secondary antibody (Product # 31400) diluted 1:40,000 in blocking buffer for at least 30 minutes at room temperature (panel B). Chemiluminescent detection was performed using SuperSignal West Pico.

IgA heavy chain was detected at ~55kD using a mouse anti-human IgA monoclonal antibody (Product # MA5-11208) diluted to a concentration of 0.5ug/ml in blocking buffer, overnight at 4C on a rocking platform, followed by an HRP-conjugated goat anti-mouse light chain secondary antibody diluted 1:40,000 for at least 30 minutes at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico. *The band at ~25kD is a result of spill over from an adjacent lane.

The membrane was probed with a Rabbit anti-Human IgD (delta-Heavy Chain) secondary antibody (Product # PA5-16400) at a dilution of 1:200 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Plus Western Blotting Substrate (Product # 32132). Results show a band at ~50-75 kDa.
