Product Specific Information
PA5-12092 targets SQSTM1 in WB, IHC and IF applications and shows reactivity with Human samples. The PA5-12092 immunogen is a synthetic peptide from the C-terminal region of SQSTM1 conjugated to KLH. PA5-12092 detects SQSTM1 which has a predicted molecular weight of approximately 48 kDa.
This gene encodes a multifunctional protein that binds ubiquitin and regulates activation of the nuclear factor kappa-B signaling pathway. The protein functions as a scaffolding/adaptor protein in concert with TNF receptor-associated factor 6 to mediate activation of NF-kB in response to upstream signals. Alternatively spliced transcript variants encoding either the same or different isoforms have been identified for this gene. Mutations in this gene result in sporadic and familial Paget disease of bone.

Immunofluorescence with anti-SQSTM1 Polyclonal Antibody (PA5-12092)
Immunofluorescent analysis of U251 cells stained with anti-SQSTM1 Polyclonal Antibody (PA5-12092). U251 cells were treated with Chloroquine (50 ?M,16h), then fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min), cells were then incubated with anti-SQSTM1 Polyclonal Antibody (PA5-12092) (1:200, 2 h at room temperature). Primary antibody detected by Alexa Fluor® 488 conjugated donkey anti-rabbit secondary antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ?g/ml, 5 min).
