The MA5-14943 immunogen is synthetic phosphopeptide corresponding to residues surrounding Thr183/Tyr185 of human SAPK/JNK.

MA5-14943 detects Phospho-SAPK/JNK (Thr183/Tyr185) which has a predicted molecular weight of approximately 46 kDa, 54 kDa.
General Information
The protein encoded by this gene is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This kinase is activated by various cell stimuli, and targets specific transcription factors, and thus mediates immediate-early gene expression in response to cell stimuli. The activation of this kinase by tumor-necrosis factor alpha (TNF-alpha) is found to be required for TNF-alpha induced apoptosis. This kinase is also involved in UV radiation induced apoptosis, which is thought to be related to cytochrom c-mediated cell death pathway. Studies of the mouse counterpart of this gene suggested that this kinase play a key role in T cell proliferation, apoptosis and differentiation. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

    Western blot analysis of Phospho-SAPK/JNK (pThr183/Tyr185) was performed by loading 20ug of THP-1 whole cell lysates, from cells either left untreated (left lane) as a control (C) or treated with 0.3M H2O2 (right lane) for 30 minutes, per well onto an SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat dry milk in TBST for 1 hour at room temperature. The membrane was probed with a Phospho-SAPK/JNK pThr183/Tyr185 monoclonal antibody (Product # MA5-14943) at a dilution of 1:1000 overnight at 4ºC, washed in TBST, and probed with an HRP-conjugated goat anti-rabbit IgG secondary antibody at a dilution of 1:40,000 for 1 hour at room temperature.     Immunoprecipitation of Phospho-SAPK/JNK (pThr183/Tyr185) was performed on HEK293T cells. Antigen-antibody complexes were formed by incubating 500ug of HEK293T whole cell lysate (in 500ul volume) with 5ul of a Phospho-SAPK/JNK pThr183/Tyr185 monoclonal antibody (Product # MA5-14943, Lane 2) or a rabbit IgG isotype control (Lane 1) overnight at 4ºC. The immune complexes were captured on 30ul of protein G agarose, washed extensively, and eluted with 6X Laemmli buffer. Samples were resolved on a 10% SDS-PAGE gel, transferred to a PVDF membrane, and blocked with 5% milk in TBST for 1 hour at room temperature. The membrane was probed with a Phospho-SAPK/JNK pThr183/Tyr185 Antibody (Product # MA5-14943) at a dilution of 1:1000 for overnight at 4ºC, washed in TBST, and probed with an HRP-conjugated mouse anti-rabbit light chain secondary antibody at a dilution of 1:40,000 for 1 hour at room temperature. Chemiluminescent detection was performed using ECL substrate. Data courtesy of the Innovators Program.
